Is the Portuguese oyster Crassostrea angulata in Southern Europe endangered by the expansion of the Pacific oyster C. gigas? ArchiMer
Batista, Frederico; Boudry, Pierre; Lapegue, Sylvie; Heurtebise, Serge; Monteiro, C. C..
The Portuguese oyster, Crassostrea angulata, was introduced from Portugal to the French Atlantic Coast in the 1860s. C. angulata quickly settled and expanded and leaded to the development of a new aquacultural industry in France. In the late 1960s, mortality associated with the detection of an iridovirus, led to the wipe out of C. angulata from French Atlantic waters and to the massive introduction of C. gigas to sustain production. In Southern Europe, similar symptoms were also observed in natural stocks of C. angulata from Sado River (Portugal) and from the area of Cadiz (Spain). Nowadays, only very few «pure» populations of C. angulata remain in southern Europe. These populations are potentially endangered by the current expansion of C. gigas...

Microsatellite analysis of 6-hour-old embryos reveals no preferential intraspecific fertilization between cupped oysters Crassostrea gigas and Crassostrea angulata ArchiMer
Huvet, Arnaud; Balabaud, K; Bierne, Nicolas; Boudry, Pierre.
Experimental examination of reproductive isolation is the first step in understanding li,hybridization processes. Here, we studied preferential fertilization between 2 cupped oyster taxa, Crassostrea angulata and Crassostrea gigas, as a potential prezygotic reproductive isolation. Early examination of sperm competition is now possible by molecular analysis of oyster embryos. This avoids the confounding effect of differential mortality during the larval stage. Six hundred embryos were sampled from 2 crosses. Three microsatellite loci were enough to determine without ambiguity the taxa of contributing sires of embryos. No evidence of preferential fertilization between gametes from the same taxa was shown. A significantly higher contribution of the C. gigas...

Molecular data highlight hybridization in squirrel monkeys (Saimiri, Cebidae) Genet. Mol. Biol.
Carneiro,Jeferson; Rodrigues-Filho,Luis Fernando da Silva; Schneider,Horacio; Sampaio,Iracilda.
Abstract Hybridization has been reported increasingly frequently in recent years, fueling the debate on its role in the evolutionary history of species. Some studies have shown that hybridization is very common in captive New World primates, and hybrid offspring have phenotypes and physiological responses distinct from those of the "pure" parents, due to gene introgression. Here we used the TA15 Alu insertion to investigate hybridization in the genus Saimiri. Our results indicate the hybridization of Saimiri boliviensis peruviensis with S. sciureus macrodon, and S. b. boliviensis with S. ustus. Unexpectedly, some hybrids of both S. boliviensis peruviensis and S. b. boliviensis were homozygous for the absence of the insertion, which indicates that the...

Molecular identification of intergenus crosses involving catfish hybrids: risks for aquaculture production Neotropical Ichthyology
Hashimoto,Diogo T.; Prado,Fernanda D.; Foresti,Fausto; Porto-Foresti,Fábio.
ABSTRACT Monitoring of the interspecific hybrid production and trade is essential for the appropriate management of these animals in fish farms. The identification of catfish hybrids by morphological analysis is unreliable, particularly of juveniles and post-F1 individuals. Therefore, in the present study, we used five molecular markers (four nuclear genes and one mitochondrial gene) to detect hybrids in the trade of pimelodid juvenile fish from different stocks purchased of five seed producers in Brazil. Samples commercialized as pintado (pure species Pseudoplatystoma corruscans ) from three fish farms were genetically identified as hybrid cachapinta (♀ P. reticulatum x ♂ P. corruscans ). In the stocks purchased as cachandiá (hybrid between ♀ P....

Morphological and genetic perspectives of hybridization in two contact zones of closely related species of Petunia (Solanaceae) in southern Brazil Acta Botanica
Teixeira,Marcelo C.; Turchetto,Caroline; Hartke,Sara; Schnitzler,Carolina K.; Freitas,Loreta B..
ABSTRACT Interspecific hybridization has several consequences for parental species, from blurring species limits to the emergence of new taxa. Petunia axillaris and P. exserta occur in sympatry in southernmost Brazil and naturally hybridize despite their different pollination syndromes and habitat requirements. We employed genetic and morphological analyses to characterize two contact zones between the species with the aim of determining the effect of interspecific hybridization. Microsatellite loci and a morphometric evaluation of the corolla shape were used to classify individuals based on their origin as pure parental or hybrids. Corolla color was used to classify individuals a priori (white, red or intermediate, for P. axillaris, P. exserta or hybrid,...

Morphological and molecular data to describe a hybrid population of the Common toad (Bufo bufo) and the Spined toad (Bufo spinosus) in western France Naturalis
Trujillo, T.; Gutiérrez-Rodríguez, J.; Arntzen, J.W.; Martínez-Solano, I..
The use of hyper-variable markers across species is often hindered by low cross-species amplification success, a reduced level of polymorphism or a high frequency of null alleles. However, optimizing sets of reliable and informative markers that can be consistently amplified and scored across taxa is key to address questions about patterns of genetic diversity and structure, hybridization and speciation. Here we present 14 newly developed microsatellite markers in the Spined toad (Bufo spinosus), assess their polymorphism in two Iberian populations and test for cross-species amplification in the closely related Common toad (Bufo bufo). We then use the 12 loci co-amplifying in both species to the study of a morphologically intermediate population (Moyaux)...

Natural hybridization between genetically differentiated populations of Crassostrea gigas and C-angulata highlighted by sequence variation in flanking regions of a microsatellite locus ArchiMer
Huvet, Arnaud; Fabioux, Caroline; Mccombie, Helen; Lapegue, Sylvie; Boudry, Pierre.
The marine environment is of special interest for studying hybridization between closely related taxa because of the high dispersal potential of planktonic larvae, such as those of most bivalve species. The oysters Crassostrea angulata and C. gigas are known to be very close genetically and entirely inter-fertile under controlled conditions. However, hybridization in the wild had not been investigated, mainly due to the lack of nuclear diagnostic markers. In the present paper, we first estimated genetic differentiation between these 2 closely related taxa using 8 microsatellite markers. Interestingly, 5 markers displayed significant differences of allele size between taxa. The subsequent sequencing of alleles of one of these microsatellites showed several...

Pre-isolation, isolation and regeneration protoplasts from leaf mesophyll of in vivo Malus domestica ‘Anna’ cv. Rev. Bras. Frutic.
El-Gioushy,Sherif Fathy Eid El-Sayed; Kareem,Abdul; Baiea,Mohamed Hemdan Mohamed.
Abstract To isolate protoplast, a pre-treatment was completed with the order to reduce and identify the phenolic contents round the year to encourage the isolation of protoplasts. Protoplasts from in vivo mesophyll leaves of apple cultivar “Anna” was isolated from 15 days old leaves by plasmolying in medium containing 90 g L-1 mannitol for half hour, then 130 g L-1 mannitol for half hour. Then using enzymatic mixture involving (1.5% cellulase + 0.5% pectianase + 1.5% Macrozyme) Prior to isolation. Anyhow, diverse factors, for example, Osmotic pressure, incubation period, sieve pore size, centrifugation period and hormonal balance was estimated using the techniques for isolation. The quantity of cells was computed as the quantity of cells per square on...

Re-examining the hypothesis of allopatric distribution of Myoprocta acouchy and M. pratti (Mammalia: Dasyproctidae) in South America Pap. Avulsos de Zool. (São Paulo)
Ramírez-Chaves,Héctor E.; Suárez-Castro,Andrés F.; Patterson,Bruce D..
Currently, two allopatric species of acouchies, genus Myoprocta (Rodentia: Dasyproctidae) are recognized. Nevertheless, there is morphological variability in the regions where the species are distributed that suggests either sympatry of two (or more) distinct species or else breakdowns in the characters that have been used to diagnose the species. We reviewed specimens of Myoprocta from Colombia and found that both reddish and greenish forms are sympatric in the Amazon basin of the country, including areas adjoining Ecuador and Peru. These records apparently refute the hypothesis of allopatry for these species in South America. However, the results of a principal components analysis showed little or no morphological separation between these two forms. In...

Reproductive behaviour of two tilapia species ( Oreochromis niloticus , Linné, 1758; Sarotherodon melanotheron, Rüppel, 1852) in freshwater intra and interspecific pairing context ArchiMer
Akian, Dieudonne; Yao, Kouakou; Clota, Frederic; Lozano, Paul; Baroiller, Jean Francois; Chatain, Beatrice; Begout, Marie-laure.
Nile tilapia Oreochromis niloticus (NT) and Black-chinned tilapia Sarotherodon melanotheron (BCT) are respectively characterized by fast growth and water salinity tolerance which attract the farmers who could take advantage of both species characteristics by producing hybrids reared in brackish or marine waters. Little information is however available about social interactions between the two species which are distinct by their mode of parental care (oral incubation). The objective of this study was to determine the behaviour of the two species, NT and BCT placed in freshwater intraspecific and interspecific pairing contexts. Females of both species (n = 20 per species) were first isolated for 90 days in a 60 L aquarium to determine the average duration...

Reproductive phenology of Melocactus (Cactaceae) species from Chapada Diamantina, Bahia, Brazil Rev. Bras. Bot.
Fonseca,Rosineide Braz Santos; Funch,Ligia Silveira; Borba,Eduardo Leite.
This paper discusses the phenological strategies of Melocactus glaucescens Buining & Brederoo, M. paucispinus G. Heimen & R. Paul, M. ernestii Vaupel and M. ×albicephalus Buining & Brederoo, species from Chapada Diamantina, northeastern Brazil. Melocactus glaucescens, M. ernestii and M. ×albicephalus occur sympatrically in an area of "caatinga"/"cerrado" vegetation, and M. paucispinus in an area of "cerrado"/"campo rupestre". The superposition of flowering in these sympatric taxa was compared and analyzed. The phenology of M. paucispinus was correlated with both abiotic and biotic factors. Flowering of M. glaucescens and M. ×albicephalus were observed to be continuous (though with moderate peaks of activity), while fruiting was...

Response to selection for growth in an interspecific hybrid between Oreochromis mossambicus and O. niloticus in two distinct environments ArchiMer
De Verdal, Hugues; Rosario, Westly; Vandeputte, Marc; Muyalde, Nerafe; Morissens, Pierre; Baroiller, Jean-francois; Chevassus, Bernard.
The development of a saline tolerant tilapia strain able to grow fast is of importance in the Philippines, where 240 000 ha of brackish water ponds are available. To this end, founder hybridization between Oreochromis niloticus (with favorable growth traits) and O. mossambicus (with favorable salinity tolerance traits) was performed and followed by backcrossing with O. mossambicus to develop a strain highly tolerant to saline environments. Genetic selection for growth performance was subsequently conducted. The aim of the present study was to estimate growth performance and survival of the hybrid in two rearing environments following four generations of genetic selection. A comparison between the hybrid and an internal reference line was performed in two...

Serological and molecular tools for strain discrimination of Citrus tristeza virus isolates from Nuevo Leon, Mexico Agrociencia
Iracheta-Cárdenas,María M.; Almeyda-León,Isidro H.; Cevik,Bayarm; Niblett,Charles L.; Lee,Richard F.; Rocha-Peña,Mario A..
Citrus tristeza virus (CTV) is an important issue to the Mexican citrus industry. CTV infected plants have been reported in 20 citrus producing states of the country as symptomless virus infections, even on plants grafted on the sensitive sour orange rootstock. The detection of severe CTV isolates is mandatory to reinforce the operation of the CTV campaign nationwide. In this study, 13 CTV isolates from Nuevo León and one from Tamaulipas states were characterized for activity against the strain discriminatory MCA13 monoclonal antibody, bi-directional (BD) PCR, and hybridization with specific DNA probes, with RT-PCR amplicons, of the p25 coat protein gene. Ten out of 13 CTV isolates from Nuevo León and the Tamaulipas isolate gave a positive reaction to the...
